Most of the Cape is sleepy during the off season, but if I had to choose I'd go with Falmouth, simply because its larger in population (more year rounders), yet close enough to the "mainland" if you needed to go off Cape for any reason (Plymouth, Providence, Boston). Falmouth has a nice little downtown area, and most of the restaurants are open off season. Falmouth also has a bike path that is pretty popular. Not sure about entertainment though.

Hyannis is centrally located and the biggest town on the Cape. It can be very busy with tourists in the summer but if you now live in a city, you will find that dealing with the extra people is no big deal. Chatham is lovely but expensive. Definitely a preppy feel. Very nice walkable town center. Orleans is a great choice too. It is the biggest town on the lower Cape.
All these towns are near beaches and outdoor areas. Cape Cod is relatively small so you can get from Hyannis to Orleans pretty quick. Hyannis has the most nightlife, all these towns have good restaurants. I went to high school in Dennis and so never went to Falmouth; we were oriented to mid and lower Cape. Falmouth is much closer to Boston if that is a consideration. Orleans is a good choice for year round living, I think.

Several of my friends' parents have retired to Harwich/Harwichport and love it. I think for them it's the best of both worlds -- scenic and a little less-traveled (at least in the off season) but not super remote and well situated between Orleans and Hyannis.

You should look into Brewster. It is one of the few towns on the cape that is not over developed. Home to the Cape Cod Natural History Museum too.

Falmouth also has the Woods Hole Oceanographic Institute so it's not just service sector people. It has a hospital. Route 28 to the Bourne Bridge (or secondary roads to the Bourne Bridge) is less of a summer disaster than the Mid Cape Highway. You can be in Boston in an hour & 15 minutes outside of rush hour. Cheap Steamship Authority ferry to MVY is an easy bike ride away.
