I think it depends on the business. First, if it says apply online, then you have to apply online, there is no getting around that. Most businesses I would say don't call or drop by, except in retail. I have witnessed managers being approached in stores by someone looking for a job. And if they have already done the application online AND the store is actually hiring the manager may grant an interview. Time permitting.

Most of the time the person has not yet applied online and are told to go do that. Many companies always have job listings just so they keep getting people into their database for future work, like around the holidays. There may not actually be a real opening.

Companies like Home Depot and Lowe's won't be able to do anything in person. Their hands are tied by the system. They can not hire anyone that does not pass through their applicant tracking system. This is for legal reasons. If they do they will be quickly fired.

I don't think it will hurt you because it is an organizational policy not a store policy, but it definitely won't help and won't get you anywhere because they can only interview people that make it through the pool and onto the manager's computer screen.
