Charlotte, DC, Atlanta, Miami, Austin, Houston, Seattle, Denver, Seattle and many others are all transforming really quickly & rapidly.

I would rank Philadelphia's downtown vibrancy (I think that is what we are referring to) as top 3-5 guaranteed. Top 2 is maybe a stretch though. Greater Center City does have the second-most densely populated downtown area after Midtown Manhattan.
New York is undisputed #1. I would rank Chicago as #2 (it's not that far ahead of PHL and SF though), and a tossup between SF and Philadelphia for a close #3/4, the pool becomes more crowded for claiming the 5th spot, but DC or Boston could make the claim.
After the top 4-6 there is a drop-off in downtown vibrancy. The cities you mentioned at the end are indeed changing rapidly, but they are still a tier down (or several) from most vibrant downtown, with the exception of DC.
