Is there any significant difference between these cities when it comes to: politics, vibe, weather, cost, yuppie-ness, outdoors opportunities? Which city would you choose?
Indianapolis has a great underground music scene, some grime and some glitter. I think Columbus is pretty bland, and Kansas City was nice enough, I just haven't been there enough times to form an opinion.

I'm not sure why people have this impression of Columbus--but it is easily the most vibrant of the three cities. Having the largest university in the country in the middle of the city keeps makes sure C-bus is anything but bland.
I don't know weather to choose between Columbus and KC, as i haven't been to them and did not like Indianapolis. Kansas City appeals to me a lot for some reason and columbus seems like a cool place. I love their LeVeque Tower!
But since you asked, I'll take a stab at your original question. On paper all three cities are fairly similar
-rougly the same size (~2,000,000)
-somewhat spread out
-similar low cost of living
-healthy growth rates
-similar political leanings
-similar weather (perhaps warmer and drier in KC)
-neutral, flat accents
but as far as the vibe is concerned, I think KC has more of a western/great plains feel to it, while Indy and Cols seem more traditional midwest/great lakes region. Indy feels to be more "rust belt" of the three and probably has the roughest central city. I believe Cols appeals to the yuppie types more, and Indy seems more touristy/family oriented. KC is the hilliest of the three.
