Go to FreeMapTools.com and you can find out how much population there is within a given radius of a specified point. For instance within a 5 mile radius from downtown...
Greenville - 134,214
Charleston - 98,398
Columbia - 138,509
If you wanna go with a 25 mile radius from downtown of each city
Greenville - 692,993
Charleston - 492,523
Columbia - 588,556
Others.....(10 mile radius)
Charlotte - 569,809
Atlanta - 957,443
Greensboro - 306,684
Raleigh - 450,636
Knoxville - 291,862
Couple of notes
- Greenville and Columbia have similar sized core populations within their city center
- for some reason Columbia shoots ahead by 30k when you get to 10 miles out
- Greenville destroys the other 2 when you get 25 miles out of the city center
- 25 mile radius for Greenville means you're butting up right to spartanburg city and right up to anderson city. so NO it does include the city populations of those communities. It would include for instance duncan, greer, lyman, roebuck, etc... western suburb populations of western spartanburg county. DOES include Easley and Pickens in Pickens county.
- Columbia's 25 mile radius does include as far north as Winnsboro, as far south as St. Matthews, as far west as Batesburg - Leesville and as far east as Lugoff

And probably the most important note, although obvious, is that Charleston is at a disadvantage here being directly on the coast.
I have no idea what Charlestons artificial population is when tourists are considered but damn its crowded here. Heck...this time of year probably no city has more people within 10 miles than Myrtle Beach.
Yep. Thats why Charleston feels denser. Its circle cut in half. And...the thousands of tourists and college kids dont count.
So you're saying college kids don't count. What about all those people at Carolina and Ft. Jackson in Columbia? I'm pretty sure Ft. Jackson counts, and that's a pretty big number.

Fort Jackson has very few permanently-stationed personnel on base, and I doubt recruiting classes are counted.
