They want experience, not knowledge.....
Probably the best way to learn it is pick up a copy of QuickBooks pro and I HIGHLY recommend "The official guide" by Leslie Cappachetti (you can find it on Amazon for 18 bucks).
You can probably pick up enough knowledge with the book and the tutorials included in the software. If you want a pretty good (and cheap) additional video course "Computeach" has a QuickBooks course on Amazon for 20 bucks or so. I have NOT viewed it but the ones I have watched were well worth it. (don't buy directly from computeach unless you want to pay five times the cost)

I have a side business working with QuickBooks/Enterprise/Point of sale mostly doing troubleshooting and reporting. That's why I made the initial comment, it seems like businesses do not TEST applicants on their knowledge of QuickBooks, they think that a year of two inputting data is more relevant than knowledge.
I don't know how to get around the experience they are looking for. You might find someone willing to give you a chance.
