Good list. I'd add to this Rio, Florence, Amsterdam and Vancouver. Also, maybe LA for the universally recognized Hollywood sign, and Las Vegas for the Strip.

PuebloFuerte-those images of Cartagena are magnificent-still need to get myself there for a long weekend! However, I do have question about which places are considered "major cities" Cartagena itself isn't really the primary commerce, etc. center in it's own region (Barranquilla).. nor is Venice. That said, I agree, both EXTREMELY distinguishable.
Here are a few that I think would warrant at very least, strong consideration.
1. Rome
2. Washington DC
3. San Francisco
4. Amsterdam
5. Moscow
6. Paris
7. New York City
8. Sydney
9. Shanghai (Hong Kong MIGHT be more on volume-but Shanghai deserves note)
10. Istanbul
I think these would be difficult to confuse for someone with general world/travel knowledge and exposure to it.
A few secondary cities that haven't been mentioned that to me are easily identifiable in most pics - and the most famous pic spot
Bangkok - Grand Palace
Buenos Aires - La Boca
Chicago - Chicago Skyline from Lake Michigan
Jerusalem - Old City from Mount of Olives
Kathmandu - Durbar Square
Marrakech - Djemaa el Fna
New Orleans - French Quarter
Vienna - Hofburg or Saint Stephen's
