I don't know, but it certainly wouldn't be anything made in the last 20 years.

Second would be Office Space.

Probably a Monty Python movie (either Life of Brian, or The Holy Grail, or The Meaning of Life), followed by Young Frankenstein and The Big Lebowski.

Not a big fan of comedies (never see them in the theater) but I always laugh at Easy Money with Rodney Dangerfield and Joe Pesci. I also think A Christmas Story (Ralphie and his red ryder bb gun) is pretty funny.

Funniest movie would be going to a movie theater playing one of the Paranormal Activity type crap horror movie with cheap scares and recording the audience muted for the duration of the movie while playing one of the Bloodhound Gang songs in the background.

Seriously though, how can anyone just pick one? 70s and 80s were arguably the best decades for comedy and I can't even pick one from those decades alone. Blazing Saddles? Animal House? Spies Like Us? See No Evil Hear No Evil? Beverly Hills Cop? Top Gun? Airplane?

Puhleezeee don't ask me to pick just one.
