Having lived in SC (Greenville) for 22 yrs.... Traveled extensively conus and overseas. The numbers are meaningless. There's still only two places in SC that any outsider identifies with....Charleston and Hilton Head. The rest take a backseat.
Hmm, seeing as Hilton Head has a city population of about 37,000 people and the smallest urban area in the state, it isn't even close to being considered a big city.
Columbia - 550,000
Charleston-North Charleston - 548,000
Greenville - 405,000
Myrtle Beach-Socastee - 215,000
Spartanburg - 180,000
Mauldin-Simpsonville - 120,000
Rock Hill - 105,000
Florence - 90,000
Anderson - 75,000
Sumter - 73,000
Hilton Head - 68,000
As itsallaboutyou said, besides population (since population is pretty much meaningless, I can agree with him/her on that), the only places that people outside the state that know on a name basis would be Hilton Head and Charleston.
For instance, I was giving my address to someone that was going to ship something to my house from Arizona. He asked what city I lived in and I said "Columbia". He replied with... Columbia... Where? What state? If I would have said Hilton Head or Charleston, that person probably would have been more apt to know automatically off the bat the Hilton Head or Charleston I was talking about since they are more renowned over the country.

In my experience people often think Charleston is in North Carolina.

Hilton Head and Charleston are not immediately recognized on a national scale. Sorry, that is just not true.
