Which is the best place to live and why? What are the positives and negatives of living in these towns?

They are hugely different. I am going to guess you have never visited these places. My advice to you - table any attempt at a decision until you have visited each place in person.

Anyway:
Santa Cruz: wealthy, liberal, mostly white, gorgeous, isolated
Santa Barbara: ultra-wealthy, moderate-to-conservative, mostly white, gorgeous, isolated
La Mesa: suburban, diverse, middle-class
