I'm in the Triangle (NC) and haven't been to Florida since I was a little kid. I'm sure we drove through Jacksonville but I don't remember it, so I'll go with Jacksonville.
I've never stopped in Greenville, SC or Spartanburg, too. I've only driven through on the way to Atlanta so I'm not sure if that counts or not.

(I've had a lot of free time on the weekends so I've seen a lot of Washington and Oregon in the few months I've been here)
By all means take in Victoria, BC when it's possible. It's a lovely city, and the island is absolutely beautiful. We enjoyed the ferry back over to Vancouver and the ride back into Seattle.
You didn't miss anything in Bloomington. South of Chicago is flat as a board and it's only beautiful to a row crop farmer. It's a sea of corn--as far as the eye can see.
From Chicago to Miami, the only major city I'm not familiar is Charlotte. I have been there for meetings, but have just driven through on I-85.
Heck, I don't even use road maps from Virginia to Florida to Texas to Missouri and back to Chicago. Product of being a mega mile traveler for work before retirement.
I'm in Austin and the closest major city that I've never visited is Monterrey, Mexico, which is about 7 hours away. The next closest is Kansas City which is 11 hours away, though I've also never been to Tulsa (7 hours away) if that counts.
In Texas I've visited the major cities in the Texas Triangle many times, and I've also been to most parts of West Texas on multiple occasions. However, I have never visited the northeast part of the state and cities like Tyler, Nacadoches, Longview, or Huntsville.

I remember that according to my 1965 World Book (long discarded) Tulsa was either the 949th or 50th largest city in the U.S. So it counts.
Currently live in Miami and if "Major City" is a Top 50 US MSA, then the closest one I've never been to is Virginia Beach-Norfolk. But there are only two others that I haven't been to: Columbus, OH and Memphis, TN, although Memphis I have been on an airport layover lol.
