I tried to use bleach and Ajax powder form solution to get ride of a black rim in the toilet, not sure what the previous owner did to the toilet, but none of these worked and I scrubbed hard too. Any suggestions on a cleaning solution? thanks.

Most hardware, home improvement stores, sell a pumice stone that will remove this, of course need some gloves,, good luck!

get a pumice stone. It is a gray looking bar, about 6 inches long, about 1 inch square. I can't think of the 'name' , maybe Scouring stick? I think you can find it at Home Depot or Wal mart.

Mineral deposit as opposed to bacteria/fungi/mold.

yes, it is build up of calcium or other deposits. You will be amazed once you get a stone.

What keeps the pumice stone from scratching the porcelain of the toilet? Seems like if it's abrasive enough to remote the ring, it's abrasive enough to do some porcelain damage.

Yes, I am sure it might eventually but I have never had that problem. Some restraint might be needed but after the initial usage once a month usually works for me, with a light go over.
