That's more of a NY, New York City question, since they're both in NYC, but I think the most inexpensive way would be a local bus from La Guardia to where you can pick up a subway to Port Authority.

I think if you go to the Port Authority website for LaGuardia,, here you go: check this site: Public Transportation - Ground Transportation - LaGuardia Airport - Port Authority of New York & New Jersey

Looks like the M60, but it goes to uptown Manhattan, as far south as 106th St,,, so somewhere you get on the subway to go down to 42nd St Port Authority Bus Terminal. There's a map on the last page of the schedule here: http://mta.info/nyct/bus/schedule/manh/m060cur.pdf

Looks like at 125th St you could get on the A or C subway direct to 42nd St.

Other possibilities in Queens but you'd have to change subways.

Cheapest would be bus/subway, they say you can transfer free. I've never done that but you could always ask the NY folks.
