Infill would/may be choppy in between Rochester and Syracuse in part due to Montezuma and even in that portion of Route 31, which be the closets main road next to the Micron facility.
Then, you have to consider that Utica already has the Wolfspeed facility up and running. So, I could see more infill or a more likely chance of infill in between that 45-50 minute stretch.
That's why I keep calling for the tolls to be removed.
UofR and RIT are mentioned in these projects, that's why I believe there may be one or two acres available to develop somewhere between Rochester and Syracuse. haha

I’m thinking more about the land in regards of the Wildlife Reserve in between around Auburn/Seneca Falls/Port Byron, etc. along the Thruway. With that said, north of the Thruway/Reserve may be an option for infill around Clyde, Savannah and Lyons.
Also, some take I-690 west to Route 370 to Route 104 to Rochester from Syracuse and Route 31 is further north. So, you can do Route 31 to 370 to 104 to Rochester.

I see plenty of land along that stretch with for sale signs. I'm thinking more Newark, Geneva, Shortsville, Canandaigua, Victor.

Those places too. I'm just thinking that the Refuge(not Reserve) would be protected, as it is a Federal property and is in between Auburn and Seneca Falls. Otherwise, that whole area would likely be up for development for sure.
Waterloo and Seneca Falls may also get some more development as well. Auburn, Weedsport, Port Byron and the previously mentioned Wayne County communities would make sense as well, if everything falls into place.

It is a pretty straightforward route that goes through the Montezuna Wildlife Reserve and by a few landmarks/towns(Del Lago Casino, Waterloo Premium Outlets, Batavia, etc). Of course you have the immediate Buffalo area before heading up to Niagara Falls as well.

In terms of Montezuma, I just mentioned it a couple of posts above, but it is a pretty big refuge of about 10,000 acres in between Seneca Falls and Auburn. Here is a street view of a couple of signs for the refuge: https://www.google.com/maps/@42.9626...7i16384!8i8192

Montezuma NWR is an important place for bird migration. It's best to think of it as a kind of northern freshwater Everglades for water moving slowly downstream between Finger Lakes watersheds and Lake Ontario.
^Yup and no way is it going to be touched in terms of development. So, that portion of the drive in between Rochester and Syracuse will not be touched.
