Other than the given...already planning on seeing Alamo, Riverwalk, and Japenese Tea Gardens.
Also, what is Mission San Jose? I heard that is a must see but don't quite understand what it is. Granted, I could look it up online, but thought I would also ask here.

Mission San Jose is one of the 5 missions in the
San Antonio Missions National Historical Park (U.S. National Park Service)
It's probably the most "touristy" of the missions and will give you the best overview of what they were about. If you like what you see there, you can always visit one of the other missions. They're each a few miles apart, mostly to the south of downtown. Mission Espada is my second-favorite mission.
The suggestion of "Stone Oak" above was offered with tongue in cheek. Stone Oak is a master-planned city within the city, basically a suburb.
If you're not into amusement parks, it sounds like you're touching the right bases of what to see in San Antonio. I always recommend that newcomers take the Riverwalk Boat Tour. You'll get more out of the visit to the Riverwalk if you get the historical spiel from the boat captains than you will just walking around and gawking at the restaurants.
You might also consider checking out the
Market Square on the western edge of downtown. It's a slice of culture that's very tourist-friendly. I'm guessing it and anything else downtown will be extra crowded this weekend with Fiesta.

Heard that Mi Tierra and La Margarita are must tries (we love Mexican food). Not into the amusement parks...more into stuff like historical, beautiful landmarks; relaxing, soothing, take-it-all-in spots; and some shopping!
I think Bo hit on pretty much what we were considering.

We're gonna stay at the Hilton Palacio del Rio, so hopefully we will enjoy some nice, relaxing time at the hotel also!

Take the downtown ghost tour. We went on the ghost hunt of San Antonio tour and had a good time. Nope we didn't see anything but we got a walking tour with lots of interesting stories. For anyone who goes on this, wear comfy shoes so you can keep up with the group!

Heard that Mi Tierra and La Margarita are must tries (we love Mexican food). Not into the amusement parks...more into stuff like historical, beautiful landmarks; relaxing, soothing, take-it-all-in spots; and some shopping!
