Hartford is out of its league with these 2 cities, unfortunately. Hartford's in bad shape for a state capital. Has some good and super nice suburban areas--with 1 or 2 decent city neighborhoods, but all in all, the crime, inner city bad neighborhoods and mostly dead downtown after 5pm is not a dynamic model for any city.
Louisville and Buffalo are interesting cities with lots of strong identity and history. Buffalo obviously a rust city that has struggled some over the past 50 years, but has some excellent culture and great neighborhoods, along with some amazing food.
Louisville is on its way up and sort of a "gem in the rough," and not really discovered yet by a lot on the national scene. I like Louisville a lot and think it has a really bright future ahead. It has a great food/bar scene and nice, slowly developing downtown area.
Buffalo and Louisville are excellent cities. I love that each has a strong community feel, are urban, but incorporate biking and green space. I would choose Louisville overall. Louisville is gradually making steps to move into the next tier of cities.
I wouldn't sleep on Hartford for arts, especially Preforming arts, Hartford Stage/the Bushnell gets lots of Pre-Broadway shows, so they get to see shows before New York gets them.

Overall Louisville has the most going for it. It draws a lot of interest for many categories, is still “up & coming”, affordable, and overall just the most stable
I think something to consider with this comparison is that Buffalo and Hartford are also within or adjacent to high population areas(Golden Horseshoe and Bos-Wash Corridor). So, that may be something that could impact things in terms of amenities and proximity to things to do.
