I live in Nassau and want to pick up an item for sale on Craigslist that is located in the Hamptons. Is traffic going to be backed up for miles because of people heading out there for the holiday?
While I agree with kayfouroh (as I usually do), there are things to consider. What time are you thinking? Friday people will be getting out early. So if you're talking about driving from 4 until 9? I'd say no. Saturday you'd probably be okay until about 5, IMO.
I'd recommend LIRR if you're going out east. But that's me.

Yeah I was (incorrectly) assuming this was going to be during a normal hour.. i.e. anywhere between 3-7/8 on Friday and morning/afternoon on Saturday.

Your best bet is to leave very early in the morning, otherwise it's going to be bad.
I can't take the train. I'm buying something that I have to haul back in my van.
Get up at 5Am and drive. You can be sitting on the beach by 6:15 or so and you can wait around for whoever you're buying from to wake up.
Or leave at 9, get there by 11:30 with 2 years taken off you life.
