Tupelo. I've been to Jackson, but not Tupelo. Outside Memphis, I just find West TN to be pretty blah. I know people who have been to Tupelo, and they raved about how fun it was to visit. I'm not really an Elvis fan, but having been to Jackson TN, I think I'll take my chances with Tupelo. I get the impression that Tupelo punches above its weight a bit whereas I don't get that feeling with Jackson.

Huntsville AL or Little Rock AR
Huntsville. Heard it's safer and has better job market. Charleston SC or Charleston WV?
Would you rather live in St George, UT, or Burlington, VT?
St. George. I'd rather be too hot in the summer than too cold in the winter. Plus, I prefer the politics of Utah to those of Vermont. That said, I'd miss the eastern greenery when living in my new desert home.
Bristol, TN or Bristol, VA? (They are legally separate cities, so I think it counts as a comparison.)

Probably the Bristol, VA side. Since it doesn't have the NASCAR race course, and the Virginia one seems to have lesser crime than the Tennessee one. To be honest, I suspect I'd be fine living in either Bristol.
The Virginia one has a casino that was recently built, and now open. So if I didn't live near the new casino(to not be near all the traffic going there), I'd be fine with the VA one.
Aiken, SC, or Phenix City, AL? (you can see the theme I'm going with for this next one, smaller towns across the river from a bigger city)

Aiken. I'd prefer that part of SC to that part of GA. Tempe AZ or Bellevue WA?

Bellevue wouldn't be my first pick in the Seattle area but I'd take it over Tempe any day. Even if you transplanted Tempe to the Seattle area with Seattle climate and nature, I'd still take Bellevue. The downtown is more built-up and interesting, Bellevue Downtown Park is a great public space, and I'd rather be bombarded with tech workers everywhere than big-state-u college kids.
The locations of two jobs a friend is interviewing with: Columbia, MO or Spokane, WA?

Columbia. Spokane is in Eastern Washington, and while it has milder winters than the Central Plains, there's not much else around it. Columbia's a college town (flagship Mizzou campus), and while I don't think it's as cool as Iowa City, Lawrence, Lincoln or Champaign-Urbana (to name the flagship college towns of the four other Midwestern states that border it), it's still pretty lively, and it's about equidistant between St. Louis and Kansas City if your friend wants a "big city" getaway. (If it matters at all to your friend, Boone County, of which Columbia is the seat, joins St. Louis City, St. Louis County and Jackson County (pre-WWII Kansas City) as blue islands in a red sea. IDK if that's a plus or a minus, but thought it might be worth noting.)
Now, as for those two cities: Kansas City or St. Louis? (I'm sure I've asked this a few times already.)

Crime aside, St Louis. Now KCMO is also a very cool at times hip city, and has great museums such as the WW1 Museum, Negro League Museum, and also great BBQ, but at the same time, there is nothing much to do outside of downtown, St Louis actually has a huge amount of amenities such as the Zoo (one of the few free ones in America), lots of museums, and of course the iconic Arch. Not to mention Blues music.

Another Midwest city duel, Detroit or Milwaukee?
Give me Milwaukee. I am impressed with Detroit's progression and improvements. But I do feel like Milwaukee is further along, plus they have such a nice lake front. Slight edge towards Milwaukee now, maybe Detroit in the future.
Flagstaff AZ or Bend OR?
